Description

Illustrate concepts in electrophoresis with this easy-to-use kit, which lets students separate colorful dyes based on electrical charge and mass. Simply put the gel comb in the chamber, microwave the gel and pour it into the chamber, then load your dyes into the wells using the supplied micropipets. After covering the dyes and gel with running buffer, connect five 9V batteries in sequence to the electrode terminals. In just 45 minutes, you'll see distinct bands of color formed in each lane, as each dye migrates a different distance toward either the positive or negative electrode. By measuring the migration distance of each dye band, students can calculate both the molecular charge and the base pair equivalent size of each fragment. Includes illustrated teacher's guide with step-by-step instructions. The Class Set includes one kit with materials for eight separations and six sets of hardware. Units are supplied with solid BD Vacutainer stoppers for variable tube sizes. Stoppers may be punched with a cork borer to fit tube gel diameters between 1mm and 7mm. Much of the platinum wire is enclosed inside a central power core for protection from breakage. The white central power core provides contrast to enhance dye band visibility. The lower buffer chamber is surrounded by a cooling jacket for efficient temperature control. An interlocking safety cover is provided on the upper reservoir.

Hunter Thin Layer Electrophoresis (HTLE) is used for high resolution two-dimensional tryptic peptide mapping and PAA (phosphoamino acid analysis). Peptide mapping provides fingerprints of proteolysed proteins and assists with differentiating proteins suspected to be encoded by the same or similar genes, preparing individual peptides for determining amino acid composition and sequence, and determining location of amino acid residues post-transitionally modified by fatty acid acylation, glycosylation, methylation, acetylation, or phosphorylation. The clamping system and inflatable nylon air bag remove excess liquid from the surface of the plate and prevent buffer from siphoning. This creates and maintains an ultra thin interface between buffer and sample cellulose matrix during high-voltage electrophoresis. Allows uniform cooling due to close and even contact with the cooling surface, providing reliable, reproducible results from a few dpm of metabolically labeled product. Digests are resolved in two dimensions, often yielding subtle and important clues about peptide composition. Because of the low reactivity of cellulose, peptide material may be recovered for secondary analysis such as determining amino acid composition and sequence or determining the presence and position of phosphoamino acid residues.
